We compared transcriptome of the allopolyploid grass Brachypodium hybridum and its ancestors Brachypodium distachyon and Brachypodium stacei. We sampled shoots and roots from each species grown at 25 degrees Celsius under 16-hour day photoperiod for 4 days after synchronized germination. We also sam .. [more]pled shoots and most young leaf blades from each species grown in a growth chamber under different temperature conditions for 3 and 15 days after transplanted, respectively. Total RNAs were extracted from each sample by using the RNeasy Plant Mini Kit (QIAGEN). Poly(A) RNAs were purified using NEBNext Poly(A) mRNA Magnetic Isolation Module (New England Biolabs). Libraries for single-end strand-specific RNA sequencing were obtained using the Ion Total RNA-Seq Kit v2 (Life Technologies). Size-selected libraries were purified using Agencourt AMPure XP (Beckman Coulter). The sequencing analysis was performed using the Ion Proton sequencer (Life Technologies) with three biological replicates. [less]
