| Construction Protocol | Cells were collected at absorbance of 0.5 at 600 nm. RNA was extraced using Qiagen RNA extraction kit according to manufacurer's instructions. Ten μg of total RNA was processed with the RiboMinus Transcriptome Isolation Kit for Yeast and Bacteria (Invitrogen) to deplete the rRNA. cDNA libraries were constructed using the SOLiD 4.0 System from Life Technologies according to the standard manufacturer's protocol. |
